Comparative Molecular Analysis of Candidatus Phytoplasma aurantifolia Isolates in Central Part of Iran Eliza Karimi karimieliza@yahoo com Date of submission January16 2013 Department of Plant Protection Isfahan University of Technology Isfahan 84156 83111 Iran Degree M Sc Language Farsi M Bahar Ph D Assoc Professor Supervisor mbahar@cc iut ac ir B Sharifnabi Ph D Professor Supervisor sharifna@cc iut ac ir Abstract Lime witches broom is an important phytoplasma disease which is first reported from neighboring countries in Persian Gulf and subsequently distributed extensively in Iran Detection of causal agent of lime witches broom Candiadatus Phytoplasma aurantifolia from fruit trees field crops vegetables ornamentals and weeds in Isfahan province during recent years indicates that Ca Phytoplasma aurantifolia is spread from host to host in central part of Iran Despite considerable prevalence of Ca Phytoplasma aurantifolia there is no sufficient information regarding the correlation between its genetic diversity host range and geographics distribution Samples of plants showing phytoplasma symptoms including lime alfalfa clover apple peach potato and tomato were collected from the region during summer and fall 2012 Phytoplasmas were detected in symptomatic plants by nested PCR using P1 P7 or P1A P7A followed by R16F2n R16R2 To classify the phytoplasma detected in the samples collected from different region PCR RFLP analysis were performed using different restriction enzyme For this purpose a 1240 bp fragment amplified in nested PCR were digested with TaqI HinI and HapII enzymes Based on comparsion of PCR RFLP profiles of the resultant fragments with reference strains the associated phytoplasmas were identified as the members of Aster yellows stolbur and peanut whitches broom groups Based on nucleotide drawing phylogenetic tree using sequence data resulted to seprate the peanut witches broom group isolates into two main groups The members of group 1A including strains MM1 RS3 AH1 AN1 App1 and PP8 which were very similar to each other were classified as Ca Phytoplasma aurantifolia In group 1 members of the subgroup B together with alfalfa strain AF2 and Iranian almond PA14 were found similar to Cactus Witches Broom Faba bean phyllody Soybean Witches Broom phytoplasma those previously classified as subgroup C of Peanut Witches broom group In this research alfafa strains E2 from ardestan and E3 from kerman were seprated from the other isolates and formed a distinct group B Based on the results of this study there was no significant correlation among the incidence rates of phytoplasma and their host range or geographical distribution Therefore it is necessary to use specific primer pairs designed from less conserved ribosomal genes to determine the situation of E2 and E3 strains in peanut witches broom group Key words Phytoplasma Ca Phytoplasma aurantifolia PCR RFLP Peanut Witches Broom
