59Fabrication and Morphological Study of Electrospun Poly vinyl alcohol Gelatin Sodium Alginate Nanofiber for Tissue Engineering Application Abdolrahman Omidinia Anarkoli r omidinia@ce iut ac ir Date of Submission 2013 8 30 Department of Chemical Engineering Isfahan University of Technology Isfahan 84156 83111 IranDegree M Sc Language FarsiSupervisor Saied Nouri Khorasani saied@cc iut ac irAbstractRecently electrospinning technique has attracted great attention as the simplest andleast expensive means in producing polymer nanofibers with diameters ranging fromnano to micro scale Alginate Alg is an abundant nontoxic nonimmunogenic andbiodegradable polymer derived from marine brown algae one of the major componentsof ECMs in human tissue Since Alg is a hydrophilic polymer Alg based materialsfacilitate uniform cell seeding and cell ingrowth into the matrix Alg is also known tobreakdown to simpler glucose type residues which is completely bioabsorbable Alg hasbeen studied extensively owing to its unique tissue compatibility and applications in tissueengineering including the regeneration of skin Cartilage bone liver and cardiac tissue Poly vinyl alcohol PVA is a semi crystalline polymer that is biocompatible and water soluble and it has a good formation ability for fibrous materials Its flexibility andtoughness are good so it is a typical synthetic polymer that is used to improve the physicalproperties through mixing with other materials that have poor physical properties PVAwith functional groups is useful in practical investigations of functional polymersbecause of its easy preparation as a bulk material films and fibers Gelatin Gel is anatural biodegradable polymer derived from collagens that contains many functional groups glycine proline glutamic acid hydroxyproline arginine alanine aspartic acid and otheramino acids Owning to its excellent biocompatibility and biodegradability properties Gelhas been widely used and studied in regards to many biomedical applications includingwound or burn dressings surgical treatments tissue engineering of bone skin and cartilage The aim of this thesis was to fabricate PVA Gel Alg nanofiber via electrospinning method First the spinnability of PVA solutions was observed Transparent solutions of PVA wereprepared by dissolving PVA in DW at about 80 C with continuous stirring for 2 3 h Inregard to the formation of continuous and uniform nanofibers observed by opticalmicroscopy photographs the solutions with concentrations of 15 applied voltage of 13 5kV flow rate of 0 6 mL hr and TCD of 13 5 cm were found to be spinnable and producesmooth and uniform nanofiber These process parameter were used for all next exprimens Pure Alg aqueous solution can t be electrospun No continues jet were formed at equal orless than 2 wt of Pure Alg solution polymer concentration while above this concentrationthe solution viscosity is too high for electrospinning process Good morphology and bead
