Effect of Seasonal Variation on Physiological Characteristics and Antioxidant Activity of Some Achillea Species Elham Zeinali e zeynali@ag Iut ac ir Saturday September 21 2013 Department of Plant Breeding Isfahan University of Technology Isfahan84156 83111 Iran Degree M Sc Language Farsi Supervisor 1 M Rahimmalek Assist Prof Supervisor mrahimmalek@cc iut ac ir 2 M R Sabzalian Assist Prof Supervisor sabzalian@cc iut ac ir Abstract The genus Achillea L yarrow is one of the important genus of Asteraceae family which comprises over 150 perennial herb species indigenous Nineteen species of Achillea have been recognized in different geographical and ecological regions of Iran In recent years the yarrow was considered as an ornamental plant because of high variation in color growing habit and resistant to biotic and abiotic stress Climatic conditions affect on the quality and quantity of active components of medicinal plants and determining of suitable season for increasing yield Therefore investigation of seasonal variation on active component of medicinal plant is important Hence ten genotype of six yarrow species millefolium A filipendulina A tenuifolia A nobilis A aucherii and A pachycephala were sown in a split plot according to a randomized complete block design with 3 replications During the growth stage in per season different traits including total phenolic content antioxidant capacity and morpho physiological ones were measured Alcoholic extract of the leaves stem and flower was extracted in different seasons Then the concentration of total phenolic was measured by Folin ciocalteu reagent and spectrophotometer methods Antioxidant capacity was evaluated by DPPH model and Ic50 index Also accumulation of hydrogen peroxide H2O2 and malondialdehyde MDA electrolyte leakage was measured in winter Analysis of variance for the morphologic traits showed significant differences p 0 01 among the genotype and difference seasons Amlitr introduced as an ornamental plant in spring and summer because of its early flowering and pink flower A nobilis was determined as cold resistant species Result of analysis of variance for oil content concentration of total phenolic and effect of antioxidant Ic50 traits showed significant differences p 0 01 among the studied genotype The best season for gaining high total phenolic and antioxidant capacity was determined in spring following by summer Result of analysis of variance for accumulation of hydrogen peroxide H2O2 and malondialdehyde MDA traits showed significant differences p 0 01 among the studied genotype Results of accumulation of hydrogen peroxide H2O2 indicated that At31 A tenuifolia and Amuk A millefolium were sensitive and resistant to cold respectively Also results of malondialdehyde MDA indicated that genotypes of Ap71 and Amuk A millefolium were sensitive and resistant to cold respectively Results of electrolyte leakage showed that A tenuifolia and A nobilis had the highest and the lowest electrolyte leakage respectively Finally results indicated that A nobilis and A aucherii were more resistant to cold stress Key words yarrow seasonal variation total phenolic antioxidant capacity Ic50 electrolyte leakage PDF created with pdfFactory trial version www pdffactory com
